Air Pollution – The Invisible Enemy of Memory and the Brain
For decades, aging and genetics have been regarded as the main culprits behind dementia. However, recent research has revealed another equally dangerous factor: air pollution.

Lewy Body Dementia – When Brain Proteins Misfold
Lewy Body Dementia (LBD) develops when alpha-synuclein, a protein involved in regulating neurotransmitter release, misfolds and aggregates into abnormal clumps called Lewy bodies.
These protein deposits accumulate in brain regions responsible for memory, cognition, and motor control, progressively impairing memory, reasoning, behavior regulation, and movement. Patients may also experience hallucinations, sleep disturbances, and executive dysfunction, symptoms often seen in both Alzheimer’s and Parkinson’s disease.
How Fine Particles Attack The Brain
Scientists at Johns Hopkins University (USA) analyzed data from 56.5 million Medicare patients between 2000 and 2014, focusing on those hospitalized for protein-related neurological damage. By linking patients’ zip codes with long-term PM2.5 exposure, they discovered that people living in highly polluted areas faced a significantly higher risk of developing LBD.
Animal studies confirmed the mechanism: PM2.5 penetrates the bloodstream and brain, triggering alpha-synuclein misfolding and Lewy body formation. These clumps spread across neural networks, damaging neurons, shrinking brain volume, and impairing cognition. Mice exposed to PM2.5 long-term showed marked declines in brain function, while those shielded from alpha-synuclein buildup remained largely unaffected.
In parallel, researchers at University College London and the Francis Crick Institute launched the RAPID (Role of Air Pollution in Dementia) project and discovered the direct presence of fine particles in human brain tissue, linking them not only to LBD but also to Alzheimer’s, cognitive decline, and other forms of dementia.
“This is an important and compelling study that deepens our understanding of how air pollution can drive neurodegenerative disease,” said Prof Charles Swanton, a co-leader of the Rapid project and deputy clinical director at the Crick.
“By linking fine-particulate matter exposure to the biology of Lewy body dementia, it provides a mechanistic bridge between environmental exposure and disease pathology. More broadly, the work underscores the urgent need to understand and mitigate the impact of air pollution on dementia and disease risk more broadly, given its enormous and growing public health burden.”
